SPRAY-DRIED CHITOSAN MICROSPHERES FOR SUSTAINED DELIVERY OF TRIFLUOPERAZINE HYDROCHLORIDE: FORMULATION AND IN VITRO EVALUATION

Objective: Sustained release systems have the potential to enhance therapeutic responses in long-term management of psychiatric disorders. In present study, cross-linked microspheres antipsychotic drug Trifluoperazine (TFP) were prepared using biodegradable polymer-chitosan and various vitro evaluations performed on microspheres. Methods: The spray drying technique was used prepare TFP-loaded chitosan Tripolyphosphate (TPP) incorporated into solutions as a cross-linking agent varying concentrations. Different like production yield, encapsulation efficiency, drug-polymer compatibility, Scanning Electron Microscopy (SEM), X-ray diffraction studies (XRD), Differential Colorimetry (DSC), particle size, zeta analysis developed formulations. Results: formulated microparticles exhibited yields ranging from 38.51 57.21% had reasonably good efficiencies (54.52-78.35%). excipient compatibility confirmed by Infrared Spectroscopy. All showed positive with mean diameter 1.45-3.61µ. SEM images revealed formation spherical particles indentations surface. XRD DSC presence an amorphous form inside profile TFP influenced considerably changing concentration polymer crosslinking formulation. (0.5%) reduced 91% 79%, when TPP increased 10% w/w 30% w/w. formulations clearly burst initial hours subsequent sustained profile. Conclusion: results this study suggest that crosslinked spray-dried could be promising method for developing long-acting delivery system intended effectively treat schizophrenia.
